- W62783132 abstract "Publisher Summary This chapter discusses the energized movement of ions and water by chloroplasts. Chloroplasts isolated from a wide variety of species by a variety of methods have been used in the studies on ion movement and conformational changes. For the most part such chloroplasts have lost their outer membranes, together with the soluble enzymes of the matrix surrounding the grana stacks, and consist of naked grana joined by membranous lamellae. The degree of disorganization of these structures varies with factors such as the extent of homogenization, osmolarity of the suspending medium, and age. Flame photometry, isotopic labeling and atomic absorption spectroscopy have been used to measure concentrations of ions and their distribution between chloroplasts and suspending media under different conditions of osmolarity and illumination. Although this is the method of choice for the measurement of rapid reactions, the ion-sensitive electrodes currently available have limited selectivity. An alternative mechanism is one in which H + transport is indirectly coupled to electron flow by interaction of H + transport with the energy transfer intermediates. Because of the sensitivity of the high energy state to structural derangement, workers have found it virtually impossible to investigate using classical techniques of enzymology, and indeed it has resisted attack for many years, a fact that has contributed to the attractiveness of a chemiosmotic hypothesis" @default.
