- W630470710 abstract "Mono-L-aspartyl chlorin e6 (ME2906), which is a effective photosensitizer for photodynamic therapy (PDT), emits fluorescence with its characteristic peak at 665nm excited by light at Soret band (415nm). Fluorescence image analysis system using laser diode system for PDT in order to excite ME2906 in tissue at Q band (664nm) was assembled. Both PDT and photodiagnosis can be performed with the system. we analyzed about correlation between the ME2906 concentration in tissue and the fluorescence intensity measured by the system. Balb/c mice implanted Meth-A fibrosarcoma subcutaneously in the right hind leg were used in the study. The fluorescence image of ME2906 in tumors and muscles of the other legs were captured at 0.25, 1, 2, 3 hours after the 5 mg/kg intravenous administration of ME2906 to each tumor-bearing mouse. Following capturing image, tissues were removed and the ME2906 concentration in tissues were measured by HPLC analysis. With the lapse of time after ME2906 administration, the fluorescence intensity decreased but contrast between tumor and muscle became higher. The ME2906 concentration in tissue and the fluorescence intensity were correlated very well (muscle: R2=0.975, tumor: R2=0.725). The relation between the ME2906 concentration and the fluorescence intensity of tumor was similar to that of muscle. This result indicate that this system is not influenced by autofluorescence because of high-wavelength excitation and fluorescence image analysis which dose not depend on variety of tissue is possible by the system." @default.
