• W63682408 abstract "Metabolic studies of FIAC were carried out in herpes simplex virus type 1 and mock infected Vero cells using HPLC purified (2-/sup 14/C)FIAC. HSV-1 infected and mock infected Vero cells were incubated with 2 ..mu..M (2-/sup 14/C)FIAC for 24 hrs. DNA purified from both viral and mock infected cells was hydrolyzed with DNAase 1 and venom phosphodiesterase. The resulting deoxynucleoside monophosphate derivatives were analyzed by HPLC (Whatman SAX column). Only FIAU, the deamination product of FIAC, was found in DNA from both HSV-1 infected and mock infected Vero cells, while FAC, FMAU and FIAC were not detected. Whereas 0.112% of Tdr in DNA from mock infected cells was substituted by FIAU 1.056% of the Tdr in viral DNA from HSV-1 infected cells was substituted by FIAU. HPLC analysis (Whatman SAX column) of medium from HSV-1 infected cell cultures showed that all (2-/sup 14/C) FIAC had been metabolized to (2-/sup 14/C)FIAU, while no (2-/sup 14/C)FIAU could be detected in the medium of mock infected cells. HPLC analysis of HC10/sub 4/ extracts of HSV-1 infected cells showed that the percentages of various phosphorylated derivatives were 10% FIAU, 76% FIAU monophosphate, 9% FIAU diphosphate and 5% FIAU triphosphate.
