- W64130004 abstract "Alkali labile sites (ALS) are defined as native DNA motifs highly sensitive toincubation in alkali solutions which produce portions of single-stranded DNA (ss-DNA)identified by alkaline single-cell gel electrophoresis or alkaline elution assays. This paperdescribes a method to produce and identify ss-DNA motifs using an alkaline solution inorder to detect the most sensitive ALS in mature human, boar, ram, stallion, donkey andcommon wombat spermatozoa. ALS maps were constructed by DNA BreakageDetection-Fluorescence In Situ Hybridization (DBD-FISH) after controlled proteindepletion and DNA denaturing protocols. The technique resulted in the reliableidentification of ALS in the sperm DNA of all species and revealed that these sites wereconsistently found within the same domains of the sperm genome. The most sensitiveregion in human and common wombat spermatozoa was located in the proximal portionof the sperm head; ALS densities in the remaining species were detected in a morecentral location. A possible structural role of ALS in relation to constitutiveheterochromatic regions as mediators of chromatin packing is proposed." @default.
