- W643444806 abstract "This chapter presents study in which a number of different plant and animal tissues have been fixed in formalin or acrolein and the RNA or DNA have been extracted with nuclease enzymes, or perchloric acid, before embedding in Epon. Nucleic acid localization is studied in the electron microscope employing uranyl acetate binding and protein and lipid distribution with osmium and permanganate binding. These methods have been applied to several cytological problems, including (1) the RNA and DNA nature of particulate fractions in rat liver nuclei, (2) the location of RNA and DNA in rat liver nucleoli, (3) the protein nature of the annular tubule in the rat liver nuclear envelope, () the protein nature of the meiotic chromosome core in helix spermatocytes, (5) the presence of RNA in aetiolated and normal maize chloroplasts, and (6) the localization of extractable lipids in cytoplasmic droplets and membranes. The chapter describes methods for radio autography with the electron microscope and their application to DNA distribution in the macronucleus of Tetrahymena." @default.
