Temulawak is one of the useful natural products from Indonesia such as antioxidant, antiinflamatory, antibacterial, antihepatotoxic, anticancer, inductor of glutathione s-transferase enzymes, and inhibitor of prostaglandin E2 production. Efficacy of temulawak as a natural products is caused by its curcuminoid compounds. Various treatments were applied to extract curcuminoid from temulawak. In this research, temulawak was extracted using Soxhlet method with acetone as solvent with temperature of 60, 70, and 80 °C for 2, 3, and 4 hours, and with sample to solvent ratio of 1:6, 1:8, and 1:10. All extraction condition used to study the effect of temperature, time, and sample to solvent ratio to curcuminoid content. This research was designed with fractional factorial design. Determination of curcuminoid content was conducted with visible spectrophotometry method and the optimum content was determined by Response surface method using software SAS 9.1. The models obtained was Content = 22.68111 + 2.30533(suhu) 0.01717(suhu) – 41.90500(waktu) + 7.10833(waktu) 2.59000(nisbah) + e with R = 99.34%. The model showed that temperature and time have quadratic effect toward curcuminoid content, while the sample to solvent ratio has a linear effect. The optimum condition of extraction was obtained at 67.27 °C, extraction time of 4 hours, and sample to solvent ratio of 1:6, with curcuminoid content of 31.87%.
