- W64936706 abstract "This chapter discusses the isolation and application of pneumocytes in the study of pulmonary metabolism. The investigation of any metabolic function in the lung is complicated by the unique anatomy and physiology of this organ. These complications are further compounded by the extensive cellular heterogeneity found in the lung, making the assignment of a biochemical event to a particular cell type very tenuous. The isolation of the type II cell offered a new dimension to the study of lung biochemistry and pathology. Removal of free lung cells was accomplished by sequential vascular perfusion and alveolar washing followed by vigorous shaking of a suspension of finely chopped tissue. Trypsin appeared to be the enzyme of choice with the grade of this enzyme ranging from fairly pure preparations. With the use of any of proteolytic enzymes, there is a potential for cell damage, with subsequent metabolic changes and loss of hormone sensitivity. More recently, centrifugal elutriation had been used in an attempt to obtain relatively pure populations of type II cells with reduced time and handling." @default.
