- W6634792 abstract "Myoporum spp. are common causes of poisoning in livestock in Australasia. The plants contain toxic sesquiterpenoid essential oils, the best known of which is ngaione. The intoxication is characterized mainly by liver necrosis, the zonal location of which in any particular animal may be either centrolobular, periportal, or midzonal. Suppression of the activity of the hepatic microsomal mixed function oxidases by pretreatment with drugs or toxins such as SKF525A or CS2 reduces the toxicity of ngaione for the male rat but reduction in the level of these enzymes by means of dietary protein depletion or adrenalectomy does not. The liver lesion observed in all cases is centrolobular necrosis. When the microsomal enzymes are enhanced by pretreatment with phenobarbitone, DDT, or high protein diets the toxicity for ngaione is not altered but the liver lesion is always periportal necrosis. In the intermediate, control state the location of the liver lesion is midzonal. Hepatic microsomal mixed function oxidative metabolism appears to be involved in toxicity of these compounds in animals, but details of the mechanism are not understood." @default.
