- W6806589 abstract "This chapter focuses on in situ hybridization (ISH) technique. Modifications of the original technique have been described that permit the detection and localization of nucleic acids within tissues from a variety of different organisms. The principle advantage of ISH over other molecular techniques is that ISH provides information about the location of the target nucleic acids within cells and/or tissues. Human papillomavirus (HPV) cannot be cultivated in vitro, so in situ hybridization is especially valuable when HPV infections are suspected. ISH is helpful in cases when the immunological diagnosis of viral infections is difficult and when there are no reliable, commercially available serologic reagents. The ability to localize a viral infection to a specific cell type and to determine the percentage of positive cells within a tissue is an attractive feature of this assay. The possibility of performing immunohistochemistry (IHC) and ISH on the same tissue section is an additional advantage in some cases. The most promising developments are those of semi-automated equipment and polymerase chain reaction amplification on the tissue section slide." @default.
