- W69744834 abstract "Pancreatic cancer is an exceptionally lethal disease with a mortality nearly equivalent to its annual incidence. Midkine (MK) is a heparin-binding growth factor and was first identified in embryonal carcinoma cells at early stages of retinoic acid-induced differentiation. There is mounting evidence that MK plays an important role in carcinogenesis-related activities such as mitogenesis, angiogenesis and anti-apoptosis. Increased MK expression has been identified in several malignancies including brain, breast and gastrointestinal cancers. The goal of our study was to identify new tumor-specific factors that promote pancreatic cancer progression or metastasis formation.We have established three primary pancreatic cancer lines from patients who underwent total pancreaticoduodenectomy for advanced pancreatic adenocarcinoma in our hospital. RNA expression profile were obtained using Affymetrix microarray and overexpression was validated by Real time PCR (RT PCR). Validation of overexpression on protein level was performed with a tissue microarray and western blot approaches. Depletion of MK was performed using small interfering RNAs (siRNA). The influence of intracellular MK depletion on cancer cell proliferation was analyzed with MTT assay.In all primary pancreatic cancer cell lines we have found a robust overexpression of MK by microarray analysis. Using RT-PCR and immunostaining procedures we have confirmed the overexpression and found MK to be highly overexpressed. The specific intracellular MK-knockdown correlated with decreased pancreatic cancer cell proliferation in vitro.Our data provide evidence that MK could serve as a possible therapy-target for this deadly disease. Using mouse-models we are currently investigating the consequences of a MK-knockdown on cancer cell proliferation and/or metastasis formation in vitro." @default.
