- W71297325 abstract "textabstractLeydig cells are present in the interstitium of the testisand are responsible for the steroid production of the testis.This production is regulated by the pituitary hormone LH andthere are indications that it is also under the influence oflocal factors.Heterogeneity of cell populations has been reported for avariety of cell types. Factors which can account for thisheterogeneity are e.g. differences in the stage of differentiationand differences in localization, whereas in other casesthe heterogeneity appeared to be related to intrinsicproperties of the cells. There are indications that thepopulation of Leydig cells present in the testis isheterogeneous too. Several authors have isolated subpopulationsof Leydig cells differing in steroidogenic activities.Besides Leydig cells the interstitium also containsmacrophages, fibroblasts, myoid cells, endothelial cells andmesenchymal cells. The macrophages are found closely associatedwith Leydig cells and have some properties in common withLeydig cells. This may give rise to a heterogeneity within thepopulation of ''Leydig differentiation of Leydig cells in the rat shows abiphasic pattern. Two generations of Leydig cells can bediscerned: the foetal generation and the adult generation. Theadult population of Leydig cells undergoes many changes duringsexual development. If this occurs asynchronously, this mayresult in a heterogeneous population of Leydig cells duringthis period. Little is known about the turnover of Leydig cellsafter maturation and it is not clear, therefore, whether and towhich extent differentiation of Leydig cells contribute to theheterogeneity of the Leydig cells present in testes from mature rats.The precise nature of the precursor cell which gives riseto the adult population of Leydig cells has not been elucidatedyet. Mesenchymal cells, fibroblasts, endothelial cells and alsomacrophages have been mentioned as possible precursor cells.The work described in this thesis concerns:1. the heterogeneity of Leydig cells2. the (dis)similarities between Leydigcells and macrophages in the testis3. the development of Leydig cellsThe heterogeneity of Leydig cells obtained from maturerats and the effect of the isolation procedure on theheterogeneity of the resulting cell preparation was evaluatedby comparing different isolation procedures (Chapter II.1.).Attempts to separate Leydig cells into different subpopulationsare described in Chapter II.2. Properties of testicularmacrophages and Leydig cells were compared using markersspecific for Leydig cells or macrophages (Chapter III)·Administration of a chemical compound Ethylene DimethaneSulfonate (EDS) causes a selective depletion of Leydig cellsfollowed by a regeneration of Leydig cells. This process ofregeneration and the role of the gonadotrophic hormones isdescribed in Chapter IV.The change of the cellular composition of the interstitialtissue as a whole and the changes in properties of Leydig cellsin the interstitium during sexual maturation have been describedin Chapter V.In Chapter VI the results are discussed with respect toheterogeneity, origin and development of Leydig cells and apossible role of the Leydig cells in immunosuppression." @default.
