- W72318141 abstract "This chapter describes a method for determining the acceptor activity for amino acid from transfer ribonucleic acid (tRNA). tRNAs transfer the activated amino acids to the growing peptide chains in the biological synthesis of proteins. They are specific for the various amino acids and differ in their structure from one species to another. tRNAs can be esterified with amino acids in the absence of the other components of the cell-free protein synthesis system with the aid of the amino-acyl-tRNA synthetases. This reaction forms the basis of this method, which is identical with or differs only slightly from the methods used in several laboratories. The method is applied for the determination of the content of a given tRNA in tRNA separation and for the investigation of the biological activity of physically, chemically, or enzymatically modified tRNA. The incorporation of radioactive amino acids into the acid-precipitable amino-acyl-tRNA is measured in that method. The chapter reviews the optical conditions of such measurements and the equipments and reagents used in the method. The content of nucleases, proteases, and contaminating amino acids in the reagents used for this method should be as low as possible. The chapter further discusses the preparation of solutions for the method, the stability of these solutions, the procedure of collection of sample, and the treatment of sample used in the method. It also highlights some other methods for determining acceptor activity for amino acids." @default.
