- W72820630 abstract "Livestock are routinely castrated to prevent various undesirable characteristics which accompany sexual maturity. In the swine industry, boars are castrated to prevent boar taint. Immunization against GnRF is an increasingly popular alternative to surgical castration. The purpose of this study was to examine the testicular effects of two anti-GnRF vaccines, both containing the same GnRF analogue-protein conjugate as the antigen, but each formulated with a different adjuvant system, referred to here as Vac1 and Vac2. Boars were treated with a priming and booster dose of either Vac1 or Vac2, or they remained untreated. Treatment with Vac2 resulted in a greater anti-GnRF antibody titer response than treatment with Vac1 ten weeks after the second vaccination. At this time, testis and epididymis were collected from 12 animals per treatment. Each testis and epididymis was weighed and detergent resistant sperm counts were collected from a subset of 6 animals per treatment. The group mean of paired testis weights for the untreated boars was 871.5g while animals treated with both Vac1 and Vac2 exhibited group mean paired testis weights that were greatly reduced at 269.8g (31% of control) and 223.1g (25.5% of control) respectively. Paired epididymis weights were also reduced in treated animals with boars exhibiting a group mean of 226.80g while Vac1 and Vac2 treated animals had group means of 73.00g and 60.48g respectively. Detergent resistant sperm counts for each group revealed that sperm production was the highest in untreated boars with lower levels in Vac1 treated animals and the lowest levels in those animals treated with Vac2. Group means of sperm/gram of testis were 35.6 million in untreated boars, 29.4 million in Vac1 treated boars (82% of control) and 19.6 million in Vac2 treated boars (55% of control). Group means of daily sperm production were 70.6x108 in untreated boars, 18.2x108 (26% of control) in Vac1 treated boars and 8.70x108 (12% of control) in Vac2 treated boars. These results indicate that both treatments reduced testicular function in boars, however, Vac2 had a greater effect than Vac1 10 weeks after the second dose, likely associated with the higher and longer lasting serological response." @default.
