- W74423617 abstract "In general, protein crystals are artificial products as only very few natural crystals are known. The first protein crystal was reported as early as 1864, but it took 96 years before the first protein structure was elucidated. During the last 20 years the emergence of recombinant protein production has increased the number of known protein structures dramatically. In future, analyses are likely to focus on the chemical details of single proteins and interaction details of protein complexes. Purity and intrinsic rigidity are basic requirements for protein crystallization. In the conventional sense, purity means absence of other macromolecules. For crystallization, however, it is of much greater importance that the protein itself is pure i.e., chemically and conformationally homogeneous. The conformational impurity in particular is very difficult to detect. The greatest purity is observed in proteins that do not require internal motions for fulfilling their function. Mobile parts may prevent crystallization by poisoning crystal growth. The same applies for inhomogeneous posttranslational modifications. When crystallization attempts are unsuccessful, the surface residues of a protein should be identified in the amino acid sequence and exchanged for others. Such crystal engineering is of particular relevance for solvent-accessible cysteines that may form undefined oligomers and aggregates. The crystallization apparatus seems to be of secondary importance to the quality of the protein used. Recent advances in automation are helpful, but robots are only practical if there are sufficient projects and raw materials to carry out daily runs. The major properties of protein crystals are discussed. The most essential parameter being internal order. The abundance of noncrystallographic symmetries is intriguing." @default.
