- W74439830 abstract "Microchimerism has been identified in a series of biologic situations, being due to clinical procedures or from physiological conditions. Blood transfusion is a procedure that can produce a mixture of donor and receptor cells. In blood transfusions, where red blood cells are the major component, microchimeric interference is due to donor leukocytes. Even blood cell components previously processed may contain a small portion of leukocytes. In some cases, the routine administration of blood-derived products may be the main cause of diseases because these hemocomponents from allogeneic donors contain leukocytes capable of surviving and expanding. Leukocytes present in bags of red blood cell concentrates used in blood transfusions may cause graft-versus-host disease (GVHD), as well as fever and, viral transmission, and may interfere in the genetic identification of the recipient. This work aimed to evaluate, at first, detection limit of microchimerism, using microssatelites (short tandem repeats STR), in mixed biological samples of controlled proportions and, later, to verify presence and duration of microchimerism in patients postblood transfusion in the Hospital de Clinicas de Porto Alegre. Analysis was performed in biological samples from three groups: DNA mixtures isolated previously to this study, blood mixtures and patients submitted to blood transfusion. All these samples were also analyzed individually. Allelic patterns were performed through PCR, using fluorescent primers, followed by capillary electrophoresis, with amplification of the following genome regions: D3S1358, D16S539, THO1, TPOX, CSF1PO, D7S820 and Amelogenin. These results in the group of DNA mixtures, microchimerism was observed up to 1:30 (or 3.3%). In the group of blood mixtures, microchimerism was identified up to 1:10 (or 10%). However, presence of microchimerism was not identified in samples collected up to 24 h from patients submitted to blood transfusion. These results suggest that the applied methodology is sensitive to identify microchimerism in biological mixtures up to the limit of 3.3%. Therefore, we can conclude that microchimerism in patients submitted to blood transfusion evaluated in this study did not reach the minimal detection limit for the methodology applied in this work." @default.
