• W76582840 abstract "The ventricular interstitium plays an essential role in normal function of the vertebrate heart. Alterations in the composition or organization of the extracellular matrix are thought to deleteriously affect heart development and function. A variety of chemical factors including angiotensin II, transforming growth factor-β, insulin-like growth factors and others have been shown to affect the expression and deposition of components of the extracellular matrix. However, the mechanisms whereby these factors modulate the function of the heart fibroblasts, the major producers of the extracellular matrix, are not well understood. The present studies were undertaken to begin to determine the effects of insulin-like growth factor on heart fibroblasts and the signaling mechanisms responsible for these effects. These studies indicate that insulin-like growth factor stimulates both collagen gene expression and proliferation by heart fibroblasts. Insulin-like growth factor treatment of fibroblasts results in the rapid activation of the mitogen-activated protein kinase and phosphatidylinositol-3 kinase pathways in these cells. Furthermore, stimulation of collagen expression by insulin-like growth factor is dependent on the phosphatidylinositol-3 kinase pathway while both pathways appear to be involved in the proliferative response. Recent work illustrates that regulation of the ventricular interstitium in vivo is dependent on dynamic interactions between diverse chemical and mechanical factors.
