- W77366277 abstract "Publisher Summary This chapter discusses the continuous culture (CTC) of human natural killer (NK)-T cell clones. NK lymphocytes, which spontaneously lyse in vitro a variety of tumor and normal target cells, have been observed among normal and pathological human PBL and in T-cell cultures. In a study described in the chapter, T cells were obtained either from continuous cultures derived from PBL with TCGF or from a mixed lymphocyte culture in which PBL were repeatedly stimulated by irradiated allogenic lymphocytes. Normal human spleen cells were used as stimulator cells. Single cloned cells were successfully recloned by using the same conditions as for primary cloning. T-cell clones with high NK activity, NK-Tc clones, were produced by isolation of either random or preselected cells from CTC or mixed lymphocyte culture (MLC). These clones proliferated for several weeks in the presence of TCGF and expressed high NK activity for at least two weeks. Not all cells expressed killing function and, in addition, recloning of 201E3 clone cells did not always generate clones with NK activity. The lytic function in NK–TC is not fixed as it occurs when lymphoid cells are immortalized into cell lines such as plasmocytomas." @default.
