Alkylating agents that are probably the most widespread chemical carcinogens in our environment, induce both in bacteria and mammalian cells, which appears to be the most efficient form of DNA repair are described in this chapter. In E.coli growing in the presence of low levels of N-methyl-N1-nitro-N-nitrosoguanidintehe, induced repair pathway lowers the mutation rate by almost four orders of magnitude. The main mutagenic lesion produced by such methylating agents is 06-methylguanine. In induced E.coli, this is rapidly repaired by a protein that apparently acts both as a demethylase and as an acceptor of the liberated methyl group. Hence, the existence of an efficient inducible form of repair should have a considerable influence on the carcinogenicity of different schedules of exposure to nitrosamines. For example, occasional large doses should be more carcinogenic than a steady rate of exposure over the same period of time; large dose rates, above the capacity of the adaptive response, should be disproportionately more carcinogenic than smaller doses. At least the second of these predictions seems to be true, for there appears to be a marked threshold effect for several nitrosamines." @default.
