- W78225521 abstract "This chapter discusses the high-resolution fluorescence and phase microscopy in conjunction with micromanipulation for in situ study of metabolism in living cells. In the study of living cells, the most useful microscopic methods have been phase contrast and fluorescence. There are two distinct spectra associated with the phenomenon of fluorescence: excitation and emission. In fluorescence microcopy, the fluorescent light emitted by the specimen is sent through a prism or grating spectrometer. This can either be done sequentially by sweeping the spectrometer, or simultaneously by the use of optical multichannel analyzer. The simultaneous method has the advantage that data is collected at the same time from all the spectral channels, greatly increasing the sensitivity. Moreover, in the study of the changes in metabolic rate of single cells because of the presence of substrates, inhibitors, and xenobiotics, it is desirable to perform injections directly into the living cells being studied. Since there is a reaction often within a few seconds, the injections must be carried out on the microscope stage itself. They are performed using a micropipette mounted on a micromanipulator." @default.
