- W78487540 abstract "This chapter provides an overview on antigen. There are two principal views of what happens during antigen presentation: the majority view and the minority view. According to the majority view, antigen presentation is divided arbitrarily into eight steps. The antigen enters the antigen-presenting cell by phagocytosis if it is large or by pinocytosis if it is small. The antigen-containing phagocytic or pinocytic vesicle pinches off from the membrane. The vesicle fuses with one or more lysosomes. Most of the antigenic protein is degraded into small fragments by acidic proteases, but some larger peptides remain undegraded. Some of the larger peptides associate physically with the major histocompatibility complex (MHC) molecules. Lysosomes containing the degradation products of the antigen empty their contents to the cell's exterior by exocytosis. MHC molecules with antigen fragments that are bound to them reach the cell surface by the same process. On the cell surface, the MHC-associated antigen fragments are recognized by the specific T cell receptor (TCR) of an appropriate T lymphocyte. According to the minority view, antigen presentation consists of only three steps: (1) the antigen binds to certain molecules on the APC surface, (2) via currents in the membrane, the molecule with the antigen bound to it is brought close to the MHC molecule and the constellation of the MHC molecule and of the antigen located nearby is recognized by the TCR.
