- W788094313 abstract "Background: Reactive oxygen species alter cell membrane permeability and cause the damage to macromolecules due to peroxidative processes. The question was stated whether peroxidative processes are infl uenced by age and metabolic rate of particular tissue in cattle. The aim of current study was the investigation of age-related changes in lipid and protein peroxidation in chosen bovine tissues. The samples of liver, kidneys, heart and muscles were chosen, based on specifi c metabolic rate, to compare the differences between different types of tissues. Materials, Methods & Results: The samples of tissues were taken from healthy cows aged between 14 and 27 months (n = 13) and female calves aged between 2 weeks and 2 months (n = 12) immediately after slaughter. Tissues were homogenized, centrifuged and the supernatant was used for further determinations. The lipid peroxidation intensity was examined by the estimation of the content of intermediates (RC-DEPPD) and end products (TBARS) of this process. The protein peroxidative processes were measured by spectrophotometric (SH groups content) and spectrofl uorimetric (Formylokinurenine and Bityrosine concentration) methods. Obtained results provided with the information that the intensity of peroxidative processes measured in present study differed signifi cantly between cows and calves. Differences were related to metabolic properties of examined tissues as well as the age of examined animals. The differences between overall mean values of TBARS showed higher values in calves in all examined tissues than in cows. Mean values of TBARS differed signifi cantly between examined samples apart from liver and kidney where values were similar. Overall mean RC-DEPPD values were signifi cantly higher in liver and muscles of cows than in calves but the opposite relationship was observed in kidneys and heart where the level was signifi cantly higher in calves than in cows. Moreover, values were similar in kidney and liver while differed signifi cantly between muscles and heart. Mean overall values of SH group content differed signifi cantly between examined samples apart from values in liver and heart. The level of -SH groups in liver and heart was higher in calves than in cows but in kidney and muscles the level was higher in cows than in calves. Bityrosine as well as formylokinurenine values differed signifi cantly between cows and calves. Moreover, formylokinurenine content was signifi cantly higher in muscles as in remaining tissues. Formylokinurenine level was lower in muscles of cows than in calves but in liver and kidney was similar in calves and cows while in heart was signifi cantly higher in calves as in cows. Bityrosine content was higher in calves than in cows in all examined tissues. Discussion: This manuscript provided with the information about bovine lipid and protein peroxidation in chosen tissues and compared the results between cows and female calves. The protein and lipid peroxidation is very important indicator of the amount of molecular lesions produced by ROS. The intensity of lipid and protein peroxidation can be related not only to age but also the level of metabolic rate and tissue specifi city. The differences between animals during ageing could be connected with ineffi cient antioxidant protection in calves and dynamic metabolism which is related to growing up processes, especially in post-mitotic tissues like muscles and heart. Moreover, older animals may accumulate lipid and protein peroxidative products within time. The knowledge about the intensity of peroxidative processes may help in appropriate antioxidative supplementation." @default.
