- W79437405 abstract "The authors have previously demonstrated that certain supplemental fatty acids (18:0, 18:1{omega}7) reduce the susceptibility of cultured porcine pulmonary artery endothelial cells (PAEC) to oxidant injury whereas others (20:3{omega}) enhance injury. They now extend those observations with two additional fatty acids, oleic acid (OA, 18:1{omega}9) and linolenic acid (LA, 18:3{omega}6). After 24 hr, both ETOH and oleic acid or linolenic acid supplemented cells were exposed to 100-{mu}M hydrogen peroxide (H{sub 2}O{sub 2}) in Hanks' Balanced Salts Solution (HBSS) or to HBSS alone for 30 min at 37C. Cell injury was assessed 2 hr after H{sub 2}O{sub 2} exposure by measuring % of total cell lactate dehydrogenase (LDH) released. As expected, H{sub 2}O{sub 2} caused significant increases in LDH release in non-supplemented control cells. In supplemented cells, however, OA protected cells from oxidant injury (ETOH/H{sub 2}O{sub 2} 83.9{+-}2.5% vs OA/H{sub 2}O{sub 2} 72.9{+-}4.0%, p < 0.05), whereas LA enhanced oxidant injury (ETOH/H{sub 2}O{sub 2} 41.9{+-}7.5% vs ETOH/LA 51.6{+-}6.3%, p<0.01). PAEC supplemented with OA were protected from H{sub 2}O{sub 2}-induced injury immediately, as well as, 48 and 72 hr after supplementation. Isotopic studies with OA demonstrate that 65% of the supplemental OA initially incorporates into the cell triglycerides, while 48 hr aftermore » supplementation, 79% of supplemental OA is found in the cell phospholipids. These findings suggest that the extent of fatty acyl unsaturation rather than the specific lipid compartment that the supplemental fatty acid partitions into determines whether supplemental fatty acids will enhance or reduce oxidant injury in cultured endothelial cells.« less" @default.
