- W80202343 abstract "Publisher Summary This chapter describes the synthesis of uniform peptide libraries and methods for physicochemical analysis. In a study described in the chapter, peptide libraries were made using an ABI model 431 peptide synthesizer. One millimole of RINK or preloaded Wang resin was loaded in a 65 ml reaction vessel. Standard FMOC, a varian amino tag, cycles were used except that an extra 8 ml volume of N -methylpyrrolidinone (NMP) was delivered to the reaction vessel for the deprotection and coupling steps. Positions containing single amino acids were triple coupled at 1 mmol, and the coupling times were doubled. Sites at which multiple amino acids were to be incorporated were double coupled at 0.8 mmol, and the coupling times were increased threefold. The solvent used for coupling and deprotection was NMP or 5 M ethylene carbonate in NMP. Mixtures of amino acids contained 19 of the 20 natural amino acids. Cysteine was eliminated to avoid disulfide bond formation skewing the data from mass analysis." @default.
