E ket megfigyelesből arra kovetkeztettunk, hogy a sejtfuzio, mint gyorsan lejatszodo folyamat, szerepet jatszhat az őssejtek terapias hatasaban. Ennek a hipotezisnek az igazolasara in vitro es in vivo kiserleteket vegeztunk, melyek főbb megallapitasai a kovetkezezők: 1, Izomszovetben, ahol a sejtfuzio elettani jelenseg, in vitro kimutathato volt, amit az őssejtek beepulnek az izomrostokba. 2, Edzesmunka hatasara letrejovő izomhipertrofia eseten az iv beadott őssejtek az izomszovetbe vandoroltak es funkcionalisan beepultek, javitva ezzel az allatok teljesitmenyet. 3, In vitro kiserletekben az őssejtek igen nagy aranyban mutattak fuzios jeleket, amennyiben egyutt tenyesztettuk azokat mas, differencialt sejtvonalakkal. | The goal of the present grant was to investigate the cellular mechanisms of stem cell transplantation. Using in vivo animal models of brain trauma we made the following observations: 1, Following brain trauma and stem cell transplantation a small percentage of the cells undergo cell fusion with the host cells. 2, Following transplantation there is a rapid improvement of the neurologic deficit in the animals, which is faster than what is expected from the differentiation and incorporation of newly formed tissues. Based on these two observations we hypothesized that cell fusion may be responsible for the therapeutic effect of stem cell transplantation. In order to test this hypothesis the following experiments were performed: 1, In in ivtro co-culture of stem cells and muscle fibers we observed fusion betwen the two cell types. This phenomenon is physiological, since hypertrophy of the muscle is a bona fide cell fusion. 2, In animal models of physical exercise we showed that systemic application of stem cells leads to improved performance and the incorporation of the cells into the muscle fibers. 3, Stem cells underwent fusion in cell culture when other differentiated cell types were present." @default.
