- W806987281 abstract "The drastic impact of infectious diseases on public health indicates the urgent need for developing new vaccines. In this study we propose a novel vaccine platform for delivery of recombinant antigens. It is based on antigen multimerization by its fusion to the measles virus nucleoprotein (N) known to auto-assemble in large-size ribonucleoparticles (RNPs) in multiple expression systems, particularly in yeast. We used whole P. Pastoris yeast as both production and delivery system of recombinant RNPs, rendering vaccine preparation technologically and economically attractive. Proof of concept of this platform was provided in a rodent malaria infection model with the circumsporozoite protein (CS) of Plasmodium, the etiologic agent of malaria. The CS protein from P. Berghei (PbCS) was selected given the availability of the stringent C57B1/6 mouse model of infection by Pb sporozoites, allowing evaluation of vaccine efficacy in vivo. Expressed in P. Pastoris, the N-PbCS fusion protein generated highly multimeric RNPs bearing PbCS on their surface and localized in peripheral cytoplasmic inclusions of yeast cells. In the absence of accessory adjuvants, immunization of C57B1/6 mice with heat-inactivated whole P. Pastoris expressing N-PbCS RNPs provided significant reduction of parasitemia following a high-dose parasite challenge. Evaluation of yeast lysates carrying escalating doses of N-PbCS RNPs revealed that protective efficiency of the vaccine candidate was PbCS dose-dependent. By combining yeasts that express relevant multimerized antigens, multivalent formulations can be designed and applied for large-scale production of low-cost vaccines against numerous infectious diseases" @default.
