- W80822490 abstract "Ethanol alters MAP kinases (MAPKs) signaling in liver. MAPKs translocate into the nucleus and serve as an important regulator of gene expression. Histone H3 phosphorylation is involved in chromatin remodeling and gene expression. Ethanol causes nuclear translocation of ERK and JNK. We have now examined the nuclear signaling of p38 MAPK and histone modification by ethanol and its metabolite acetaldehyde in rat hepatocytes. Primary cultured hepatocytes were treated with ethanol (100 mM) or acetaldehyde (5 mM) for different times (0–24 h) and subcellular fractions (nucleus and cytosol) were prepared for immunoblot analysis. Both ethanol and acetaldehyde increased phosphorylation and the level of p38 MAPK protein in the nuclear fraction with peak activation at 24 h by ethanol and at 30 min by acetaldehyde. These responses were ethanol (50–200 mM) and acetaldehyde (0.5–5 mM) dose dependent. Interestingly, the phosphorylation of p38 MAPK was not detected in the cytosolic fraction suggesting a subcellular selectivity of p38 MAPK signaling. Ethanol and acetaldehyde increased phosphorylation of histone H3 at both serine 10 and serine 28. SB203580, an inhibitor of p38 MAPK, blocked acetaldehyde induced phosphorylation of histone H3 by about 90 %. In contrast, no inhibition of histone H3 phosphorylation was observed by the JNK inhibitor, SP600125. These results suggest that ethanol and acetaldehyde selectively modulate nuclear p38 MAPK signaling and that histone H3 phosphorylation at serine 10 and serine 28 is regulated by p38 MAPK (Supported by NIH grant #AA11962)." @default.
