- W80919059 abstract "This chapter discusses a study to analyze whether post-synaptic depolarization is a necessary condition for the development of long-term potentiation (LTP). In an attempt to detect postsynaptic changes involved in the initiation of LTP, this study recorded intracellularly from CA1 pyramidal cells in hippocampal slices during series of tetani with and without subsequent LTP development. During repetitive trains of tetani to dendritic synapses, the excitatory postsynaptic potentials (EPSPs) summated to a depolarizing plateau. The amplitude of the plateau was determined by the frequency and duration of the tetanus, set by the degree of EPSP facilitation and depression. Normally, the frequencies of around 100 Hz gave the best sustained depolarization. The depolarizing plateau was coupled to a dendritic DC-shift shown by simultaneous extracellular recording from the synaptic layer. During five repeated trains, an additional slow depolarization developed during the late part of the plateau. Only when this was present did LTP develop. The degree of LTP was assessed by the shape of the input/output curve. The most impressive changes were the increased probability of discharge and the reduction in spike latency. An increased EPSP was less pronounced and in many cells, it was unchanged or even slightly reduced." @default.
