However, less is known about how the information conveyed by cytokines is processed at central levels. Here, we discuss evidence indicating that several of the effects of proinflammatory cytokines exerted in a “healthy” brain are amplified in the CNS due to the fact that (a) peripheral cytokines, such as IL-1, have the capacity to elicit their own synthesis in the brain; and (b) a sustained increase in neuronal activity also induces the production of physiologically significant amounts of these mediators. The relevance of brain-borne interleukin (IL)-1 and IL-6, when their production is not the result of pathological events in the CNS, is discussed taking as example their effect on the stimulation of the HPA axis, and their involvement in physiologic brain mechanisms such as synaptic plasticity and memory formation, and the control of glucose homeostasis." @default.
