- W81647650 abstract "ABSTRACT Damaged DNA regions can be selectively recognized by the tripeptide Lys-Trp-Lys and by the protein coded for by gene 32 of phage T4. Four types of chemical modifications were produced in DNA: pyrimidine dimers induced by UV irradiation; methylation of purines by dimethylsulfate followed by depurination; reaction with aminofluorene derivatives known to form covalent bonds with the C (8) and NH2(2) sites on guanine. In all cases the chemical modification alters the double helical structure of DNA. It is shown that the ability of the tripeptide Lys-Trp-Lys to insert its tryptophyl residue in between bases (stacking interaction) is strongly enhanced in chemically-damaged DNA as a result of local distortion of the double helix. The extent of stacking depends on the nature of the defect. The single-strand-binding protein coded for by gene 32 of phage T4 is also shown to recognize defects in the DNA double helix structure. These results are discussed with respect to the recognition of chemically-modified DNA by proteins involved in DNA repair." @default.
