- W81731857 abstract "This chapter discusses the technique of HLA typing by complement-dependent lympholysis. The detection of the HLA antigens carried on the cell surface of a variety of human cells may be performed by several techniques. Complement-dependent lympholysis has gained widespread acceptance as a reproducible, convenient means of histocompatibility typing in man. Human blood lymphocytes are separated from heparinized peripheral blood. The separated lymphocytes are incubated in the wells of plastic trays with typing alloantisera containing anti-HLA antibodies of defined specificity. In each test, a battery of different antisera including monospecific and oligospecific reagents is used. These sera are obtained from multiparous women and from planned immunization, and their specificity has been previously determined by comparing their reaction pattern with that of defined antisera and by analyzing the pattern of segregation within families. Complement is added to the cell–antisera mixture and incubation is continued. After incubation with complement, eosin solution is added to allow identification of viable cells that exclude the dye. The preparation is then fixed by adding formaldehyde solution. The HLA phenotype of the individual is established by comparing the pattern of positive cytotoxicity reactions with the specificities of the antisera in the positive wells." @default.
