This has made available a direct nondestructive assay for metabolite transitions, which involve changes in the concentration of this critical control substance. Control phenomena in sequential enzymes are of general interest, and control mechanisms in ascites tumor cells afford unusual experimental material for the demonstration of metabolic control principles. In an experiment described in the chapter, a detailed kinetic analysis of the changes of nucleotides accompanying the activation of respiration on adding glucose to an aerobic suspension of ascites tumor cells was indicated. Phosphate control following the addition of glucose to iodoacetate inhibited cells, or to 2-deoxyglucose (2-DOG) to uninhibited cells, could be observed, although the relationships of ATP and ADP were obscured by the loss of nucleotides about a minute after adding 2-DOG. In suspensions of ascites tumor cells, mitochondrial regulation of the intracellular ADP level allows stimulation of respiration by glucose." @default.
