- W82748573 abstract "This chapter deals with Macrophage colony stimulating factor (M-CSF), which is a survival factor for cells expressing the c-fms CSF-1 receptor that include macrophages as well as osteoclasts and immature dendritic cells (DCs). Interaction of colony stimulating factor-1 (CSF-1) or (M-CSF) with its receptor is associated with important biological effects. Human M-CSF exists as an 85-kDa homodimer. In murine cells, only a 552-amino-acid form encoded by 1.4 and 4.5 kb messages has been found. Liver expresses a 1.4-kb species, heart and lung express the 4.5-kb species, and brain expresses high levels of both transcripts, while intestine lacked detectable transcripts. The CSF-1 gene is present as a single copy in the mouse genome. All forms of M-CSF have a 32-amino-acid signal peptide, and a 23-amino-acid hydrophobic trans-membrane domain. M-CSF is a prepropolypeptide, with processing occurring at both the amino- and carboxy-termini. Only the first seven of nine cysteines in the mature molecule are required for biological activity. Glycosylation found in the native protein is not important for the biological activity. The M-CSF gene and the gene encoding its receptor are tightly linked on the long arm of chromosome 5, and the syntenic murine chromosome 11. GM-CSF, IL-3, ECGF, and the receptor for PDGF are also found in this region. Up to 20,000 receptors per cell exist for the mature M-CSF. Most recent M-CSF has been identified important in the biology of several neoplasms, notably ovarian carcinoma, and as a possible therapeutic agent in fungal infections." @default.
