- W837528660 abstract "Several approaches towards the optimization of amperometric biosensors using a soluble mediator in combination with reticulated vitreous carbon (RVC) were studied. RVC material was used as the working electrode with biomolecules (enzyme/co-enzyme) immobilised on its surface and intergrated with a FIA system. By using the soluble electron transfer mediator Meldola Blue the oxidation potentials for the electrochemical reactions were reduced, decreasing the interference by compounds that may be oxidised at higher potentials. Electrodes for the determination of glucose, lactate, acetylthiocholine and paraoxon were prepared. The influences of various parameters on the electrode sensitivity were investigated. In a glucose sensor, ferrocene was used as electron transfer mediator together with a conducting polymer (polypyrrole) for electron transport between the enzyme (glucose oxidase) and the electrode surface (RVC). The glucose oxidation was monitored by calorimetry while the system was electrochemically regenerated. The metabolic activity of yeast cells could be monitord by a DC resistometric method using the conducting polymer polypyrrole for immobilisation of the cells. The cobination of superporous agarose gel with electroactive material was used for construction of a chromatographic support material that was both electrically conducting and electrochemically active. A specially designed chromatography column containing this material was connected to a potentiostat and, by changing the applied potential, reduction or oxidation of the support material could be accomplished which influenced the chromatographic properties substantially.
