- W89090045 abstract "The elucidation of vitamin D metabolism to 25-hydroxyvitamin D3 (25-OH-D3), and subsequently to at least one hormone, 1,25-dihydroxy- vitamin D3 (1,25-(OH)2D3), has prompted interest in efficient syntheses of these 9,10-secosterols. This interest has been intensified because of the therapeutic utility of 25-OH-D3 and l,25-(OH)2D3 in treating renal osteodystrophy, hypoparathyroidism, certain types of pseudo-hypopara-thyroidism, possibly glucocorticoid induced osteoporosis, and by the anticipated effectiveness of these agents in treating senile osteoporosis. A need also exists for analogs with enhanced therapeutic ratios and limited sites and modes of action, e.g., intestinal calcium transport versus bone calcium mobilization. Besides their potential commercial value, there is an ever growing requirement in basic research for synthetic versions of well-known and newly discovered metabolites. Recent research has crystallized the realization that 25-OH-D3 is converted to a myriad of metabolites other than 1,25-(OH)2D3. Some of these may be important in their own right. This research would have been much more difficult, perhaps unapproachable, without sufficient quantities of substrates, and in the absence of synthetic versions of putative new metabolites. In this regard, progress in vitamin D research has been greatly accelerated by readily available vitamin D metabolites stemming from the contributions of synthetic organic chemists." @default.
