- W89347795 abstract "Publisher Summary This chapter discusses pressure homogenization of mammalian cells. Most of the procedures for isolating the subcellular components of cells make use of some form of density-gradient centrifugation. The development of the zonal ultracentrifuge has meant that such separations can be done with much larger amounts of material. There are at present a variety of instruments in general use for the preparation of tissue homogenates. Of the mechanically-driven types, the Potter–Elvehjem homogenizer appears to cause least damage to the cellular components. Suspensions of single cells seem particularly difficult to homogenize. The efficiency of the Potter–Elvehjem and Dounce type of homogenizers in breaking a sufficient proportion of the cells within a reasonable time is quite low. It diminishes markedly with volumes greater than 10 ml. Prolongation of the homogenization time means that those components that are liberated early in the procedure are exposed to shear forces for a longer time." @default.
