- W89813019 abstract "Since the original report of Powell & Slater (38) describing the pharmacological properties of dichloroiso-proterenol (DCI), a large number of β-adrenoreceptor blocking drugs have been synthesized in the last two decades (13,17, 34,35,41–44). Although originally developed for a limited clinical application, their introduction in medical therapeutics has truly revolutionized the treatment of almost all cardiovascular as well as many other diseases. Success of these drugs can be easily recognized by continued interest in synthesis and clinical evaluation of numerous drugs within this broad group. Although only a limited number of β-blockers are currently approved by the FDA for general use, a large number of such agents are under investigation within this country and many more are freely available in other parts of the world. Thus, a physician is often baffled as to the drug of choice from such a large number of agents and such a decision should be based upon a rational and objective considerations of pharmacological properties such as receptor specificity, selectivity and ancillary effects. Drugs are generally useful for a specified clinical condition, but it is well known in clinical medicine that all drugs possess a primary effect which forms the basis of their main therapeutic indication, and secondary effects which are either the cause of their side effects or are clinically irrelevant. The β-adrenoreceptor blocking drugs are no exception to this rule. In this chapter, the relevance of some of these factors in clinical therapy with the β-adrenoreceptor blocking drugs will be reviewed." @default.
