- W91787095 abstract "This chapter discusses the specificity of fresh and activated human cytotoxic lymphocyte. In a study described in the chapter, lymphocytes cultivated in MLC and in mixed cultures with autologous tumor cells lysed the patients' tumor cells. The highest frequency of positive experiments was obtained after autologous tumor cells (ATS) activation. Allogeneic tumor cells were lysed by lymphocytes treated with interferon IFN-α and as third party targets by MLC lymphocytes. In the ATS cultures, no allo-tumor lysis was generated. K562, the prototype natural killer (NK) target was lysed by lymphocytes of MLC and ATS. When different targets are killed by the same lymphocyte population, the effects can still be antigen specific if they are exerted by different sets of effectors in each case. Enlargement of the specific clone results in cytotoxicity against cells that carry the stimulating antigen and cells which carry cross-reactive antigens. Targets unrelated to the stimulus may be killed if the relevant clones are sufficiently enlarged. In contrast to the auto-tumor cytotoxicity, allo-tumor lysis by IFN-treated freshly separated lymphocytes was inhibited by the identical tumor cells and by the Con-A blasts from the same donor." @default.
