- W91910630 abstract "This chapter discusses the application of fluorometry as an investigative tool for vitamins, coenzymes, and their metabolites analysis. A simple procedure for the spectrofluorometric assay of vitamin A in blood following solvent extraction was introduced by Kahan. Drujan et al published another simple fluorometric procedure for the assay of vitamin A in tissue homogenates and in fasting human plasma. The chapter presents a procedure for thiamine and cocarboxylase analysis in which chromatography is used to measure thiamine, thiamine phosphate, and thiamine pjrrophosphate in bacterial extracts and enzyme incubation mixtures. It is discussed that riboflavin and flavin coenzymes have intense fluorescence, therefore, they can readily be assayed at very high dilutions. Fluorescence and phosphorescence have also been used in elucidating structures of coenzymes, mechanisms of enzymecoenzyme interaction, and mechanisms of photochemical reactions. At present, it is apparent that the luminescence characteristics of the flavins are very sensitive to solvents, solutes, oxygen, and light. The chapter discusses that pyridine nucleotides can be made to fluoresce in several ways: (1) treatment of oxidized nucleotides with alkali, (2) condensation with ketones, and (3) conversion to reduced pyridine nucleotides." @default.
