- W9199516 abstract "ABSTRACT Tyrosine hydroxylase was measured in dopaminergic nerve terminals in three different experimental conditions. Repeated injections of d-methamphetamine or dopamine agonists to adult male Wistar rats produced a progressive enhancement in stereotyped behavior. A subthreshold dose of the dopamine agonists produced a marked stereotyped behavior after a drug-free period of 2–10 weeks following repeated treatments with dopamine agonists. Tyrosine hydroxylase was increased in the mesolimbic area of the rats which showed behavioral supersensitivity to dopamine agonists. In the second experiment, tyrosine hydroxylase activity was measured in brain areas of rats after long-term isolation. In isolated rats, the Vmax of tyrosine hydroxylase activity in frontal cortex for L-tyrosine was increased without an alteration in Km values. In other brain areas, for example, striatum, olfactory tubercle, N. accumbens, there were no changes in enzyme activity between isolated and grouped rats. Tyrosine hydroxylase in superior cervical ganglion of isolated rats was increased. The enzyme in this area was found to have two distinct Km values for tetrahydrobiopterin. In the third experiment, tyrosine hydroxylase activity was measured to have significantly increased in the caudate and putamen of post-mortem schizophrenic brains. In a patient who was highly predisposed to schizophrenia and had not taken antipsychotic drugs for 3 months prior to death, tyrosine hydroxylase activity in dopaminergic nerve terminals was nearly ten times as high as that of controls." @default.
