- W92822846 abstract "This chapter describes the basic components of virus particles and various factors that are considered during virus purification. There are no generally applicable rules for virus purification. Procedures that are effective for one virus may not work with another apparently similar virus. Even different strains of the same virus may require different procedures for effective isolation. A virus preparation is pure for a particular purpose if the impurities or variations in the particles present do not affect the particular properties being studied or can be taken account of in the experiment. Important factors for the successful isolation of a virus are (1) choice of host plant species and conditions for propagation that will maximize virus replication and minimize the formation of interfering substances and (2) an extraction medium that will protect the virus from inactivation or irreversible aggregation. The particles of most small geometric viruses contain a single kind of coat protein, which range in size from about 17 kDa to 60 kDa. Plant virus families and genera with large particles— such as rhabdoviruses, reoviruses, Caulimoviridae , and tospoviruses—contain more than one kind of protein, some of which are enzymes while others have a structural role. The two kinds of enveloped plant viruses (rhabdoviruses and tospoviruses) contain lipid as part of an outer-bounding lipoprotein envelope, which is probably derived from a host bilayer membrane. All viruses contain water, with enveloped viruses containing more water than geometric ones. Many viruses contain divalent metal ions, especially Ca 2+ , and a few contain polyamines. These components may be important for the stability of the intact virus particle." @default.
