- W932342977 abstract "The aim of this study was to evaluate the technical, toxic and therapeutic aspects of autologous bone marrow transplantation (BMT) in dogs with lymphoma, as well as the effect of freezing of bone marrow bags on nucleated cell count, CD34 stem cell count and cell viability. Seven dogs with lymphoma were submitted to autologous bone marrow transplantation after eight or nine chemotherapy sessions related to the remission induction phase of the Madison-Wisconsin protocol and were followed for six to 18 months after BMT. The transplantation protocol consisted of bone marrow harvesting (10 mL/kg), followed by administration of cyclophosphamide (400 or 500 mg/m) and intravenous reinfusion of bone marrow. Bags containing bone marrow were processed in steps including red cell depletion, plasma depletion and cryoprotectors addition. Bags were stored at -80°C for four days. Freezing neither modified nucleated cells/kg and CD34 cells/kg or viability, suggesting that freezing technical procedures were efficacious. The mean segmented neutrophil nadir was 506 cells/μL and occurred three to seven days after BMT. Eleven days after BMT all dogs had normal leukocyte and neutrophil counts. Complete remission was observed in three animals followed for six, 17 and 18 months. One animal showed complete remission for six months followed by relapse and death and three animals showed relapse after 12, 40 and 87 days after transplantation. Autologous BMT was minimally toxic for the animals and supported the myelosuppression caused by high dose cyclophosphamide.
