- W935652212 abstract "Automated conversion of anilinothiazolinones into phenylthiohydantoin (PTH)-derivatives requires changes in the design of instruments—namely, solid-phase sequencer and liquid-phase sequencer. The principle of operation, however, remains very similar. Converters for solid-phase sequencing are of reaction cell type. The converter described in the chapter can be incorporated into almost any solid-phase sequencer and requires no additional programming capacity for operation. Converters for liquid-phase sequencing consist of a jacketed pear-shaped glass reaction vessel 80-100 ml in volume. The thiazolinone, dissolved in chlorobutane, is transferred from the cup into the conversion flask and the solvent is then evaporated under a stream of N2. Aqueous trifluoroacetic acid is added and conversion takes place at 55 ° for 20-30 min. The acid is removed by a combination of blowing N2 and vacuum and the PTH-derivative transferred to the fraction collector in a stream of dichloroethane : methanol (7 : 3) is ready for identification. Results using this converter were found to be impressive; however, considerable modifications were also made to the sequencer and the number of these modifications necessary for efficient operation of the converter is not yet clear." @default.
