The three cellular DNA polymerases are essentially named in the order of their discovery. The chapter also summarizes some relevant characteristics of the three cellular DNA polymerases obtained from various sources and of well characterized viral-induced DNA polymerases. The understanding of the physiological role of each of the cellular DNA polymerases can help in unraveling of the complex events that accompany and control the synthesis of nuclear DNA. Essentially, DNA polymerase α has a key role in the replication of nuclear DNA and in the synthesis of the DNA of the viruses SV40, polyoma, and adenovirus. DNA polymerase α is regarded the major replicative polymerase in mammalian cells. DNA polymerase β is the smallest of the known eukaryotic DNA polymerases and shows remarkable chemical stability under various conditions. Of the three cellular DNA polymerases, only DNA polymerase γ is capable of synthesizing continuous long DNA chains in a processive manner. HSV-induced DNA polymerase is required for synthesis of the viral DNA. In addition, almost all other members of the herpes group induce a new DNA polymerase in host cells after infection." @default.
