Passend dazu zeigten vorangegangene Experimente in Prof. Stockinger’s Labor, dass CD147Knockdown T-Zellen erhoht Interleukin-2 (IL-2) produzieren, und eine Tendenz zur T-Helfer 1 (Th1) Zytokin-Antwort aufweisen.In dieser Studie untersuchte ich, welche strukturelle Determinanten von CD147 die antagonistische Funktion von CD147 in der T-Zell Aktivierung ausfuhren. Mittels Rekonstituierung von CD147 Knockdown Jurkat T-Zellen mit siRNA resistenten CD147 Deletions- und Swap- Mutanten, stellten wir fest, dass die extrazellulare und die transmembraneDomane von CD147 kooperieren, um die IL-2 Promotor Aktivitat infolge der T-Zell Aktivierung zu verringern. Dieser Effekt war zumindest teilweise von der negativen Regulation des „nuklearen Faktor aktivierter T-Zellen“ (NFAT) abhangig. Weitere Experimente zeigten, dass auch die Aktivitat des „nuklearen Faktors k B“ (NF-kB) und die Aktivitat der „fokalenAdhasionskinase“ (FAK) durch CD147 beeinflusst werden.Da die CD147 Mutante ohne cytoplasmatischer Domane die IL-2Promotor Aktivitat verringern konnte, ubt CD147 seine Funktionvermutlich uber ein anderes Transmembranprotein aus. Deshalb wurde der Beitrag potenzieller CD147 Interaktionspartner zur Regulation der IL-2 Promotor Aktivitat analysiert. Die Oberflachenexpression von CD43 und CD98, und uberraschender Weise auch die des co-stimulatorischen Molekuls CD28, war in CD147 Knockdown Zellen erhoht. Deshalb wurden CD28, CD43 und CD98 Knockdown Zellen hergestellt, und deren IL-2Promotor Aktivitat getestet. CD28 und CD98, nicht aber CD43, Knockdown Zellen wiesen eine verringerte transkriptionelle Aktivitat des IL-2 Promotors auf. Daher konnte die Deregulation dieser Molekule fur die verstarkte IL-2 Produktion in CD147 Knockdown verantwortlich sein. Wir spekulieren, dass CD147 die Funktion dieser Transmembranproteine wahrend der T-Zell Aktivierung beeinflusst und dabei die Immunantwort verringert." @default.
