- W938889701 abstract "Interferon-γ (IFN-γ) is distinct from IFN-α and IFN-β on account of its biological properties, that is, inducing stimuli, types of cells producing it, sensitivity to acidic pH, and antigenic characteristics. IFN-γ, a product of NK cells and of activated T lymphocytes, was first characterized by its antiviral and antiproliferative activities. Several in vivo experiments support the hypothesis that an IFN-γ antagonist may have therapeutic applications in autoimmune diseases, hypersensitivities, and alloreactions. IFN-γ exerts its biological activity through the binding to a single-chain cell surface receptor. In a study described in the chapter, cDNAs encoding the extracellular domain of human and mouse IFN-γ were engineered and expressed as soluble receptors in Spodoptera frugiperda insect cells infected with recombinant baculoviruses. The protein that corresponds to the external domain of mouse IFN-γ receptor is characterized and used in vivo as a prototype of the IFN-γ antagonist. This protein does not show any strong antigenicity after in vivo injection in mice. Despite a blood half-life of only 1–3 hr as demonstrated in pharmacokinetic experiments, the mouse soluble IFN- γR was able to modify the onset of acute GVHD (alloreaction) and chronic GVHD (lupuslike disease)." @default.
