- W939231748 abstract "This chapter discusses general considerations concerning the isolation of chloroplasts and chromoplasts from Capsicum fruits and their use for three specific purposes—namely, the characterization of the phytoene synthase component enzymes, their immunochemistry, and their biogenesis. In plants, the synthesis of phytoene, the first C40 carotenoid, requires the coordinated activity of the different component enzymes of the phytoene synthase complex, which catalyze the sequence of reactions presented in the chapter. The entire pathway is readily demonstrated in vitro using isolated plastids. However, despite the conservation of this pathway, isolated nongreen plastids are by far more active than chloroplasts. The Km values for geranylgeranyl pyrophosphate (PP) and prephytoene PP are 0.30 and 0.27 μM, respectively. The activity of the enzyme is strictly dependent on Mn2+. The enzyme is sensitive to arginine-directed reagents. The different component enzymes of the phytoene synthase complex are used to elicit immune responses in rabbits according to standard protocols. The different polyclonal antibodies obtained under these conditions are not active site directed, because they fail to inhibit in vitro the enzymatic activity directly." @default.
