- W945757042 abstract "Publisher Summary The chapter presents a study on leukotriene B 4 biosynthesis by erythrocyte-neutrophil interactions. Experiments on transcellular biosynthesis should conform to the following criteria: (1) are the cells under investigation likely to occur together in vivo ; (2) are the ratios of the respective cells likely to occur together in vivo ; (3) are the cells sufficiently pure to provide unambiguous conclusions regarding the origin of the eicosanoid under study; and (4) do transformations of synthetic substrate and cellularly donated substrate correspond. The chapter outlines the experimental procedures to assess these criteria for investigations on erythrocyte-neutrophil interactions. The original investigations demonstrated that combinations of human neutrophils plus erythrocytes, with appropriate stimulation, produce more leukotriene B 4 (LTB 4 ) than neutrophil suspensions alone. This was significant because erythrocytes do not contain detectable phospholipase A 2 or 5-1ipoxygenase activity and have therefore been regarded physiologically inert in terms of leukotriene formation. In the procedure for isolation of human erythrocytes, human venous blood (from drug-free volunteers) is collected using sterile sodium citrate as an anticoagulant (9: 1, v/v). In the process of neutrophil-erythrocyte leukotriene biosynthesis, the transcellular metabolism of neutrophil-derived LTA 4 is initiated by stimulating various neutrophil/erythrocyte mixtures with the calcium ionophore A23187." @default.
