- W945964554 abstract "This chapter discusses the chloroplast cytochromes f, b-559, and b6. Higher plant chloroplast particles contain three tightly bound cytochromes which in reduced form each exhibit a characteristic α-absorption maximum: cytochrome f (553.5 nm), cytochrome b-559 (559 nm), and cytochrome b6 (563 nm). Like many other tightly bound membrane components throughout biology, they are not extracted by sonication in salt solutions, with or without prior delipidation of their parent membrane. The procedures discussed in this chapter achieve a disc-electrophoretically homogeneous preparation of each of the cytochromes with good recovery and retention of known biochemical properties. Of all the three procedures, step 1 of the b-559 and b6 procedures and step 3 are of possible value for purifying other membrane components. Cytochromes f, b-559, and b6 are shown to be intrinsically aggregatable species. Thus, a rationale for the procedures is that aqueous nonionic detergent solution maintains both the desired components and their impurities as separate monomolecular species, thus facilitating their purification. Cytochrome b6 is reduced by dithionite (Na2S204) only; b-559 and f are reduced by either ascorbate or dithionite. Assay of each component at its α maximum is performed by difference spectrophotometry with sample in both cuvettes.
