- W95200724 abstract "The tissue-fixed macrophage has been identified as a central regulator controlling the activation and propagation of the inflammatory process. Circulating monocytes localize to areas of inflammation or infection or become residents of primary tissue beds where foreign antigens are frequently encountered. Examples include Kupffer cells in the liver and alveolar macrophages in the lung. These monocytes then differentiate to become tissue-fixed macrophages. The macrophage is a component of the local response to infection, serving a key role as a scavenger of bacteria and bacterial products, an antigen-presenting cell, and a coordinator of the inflammatory response via the production of a plethora of cytokines and chemokines. These mediators result in the recruitment and activation of all other components of the inflammatory response, particularly the neutrophil. In addition, activated macrophages produce a number of products, including tissue factor and prostanoids that have profound effects on the microcirculation, resulting in microvascular thrombosis and local ischemia. Production of toxic reactive oxygen intermediates and proteases by both the macrophage and the neutrophil can result in endothelial cell injury and subsequent capillary leak. Although these processes are necessary to control and contain a local focus of infection, systemic activation of these processes can result in extensive tissue injury and subsequent organ failure." @default.
