- W95255602 abstract "Lysosomes are the major intracellular site for the degradation of a variety of macromolecules, including proteins, carbohydrates, nucleic acids, and lipids, that are internalized from the extracellular space by endocytosis, delivered by fusion with phagosomes, or derived from the biosynthetic pathway. Lysosomes are enriched with various acid hydrolases that are separated from other cytoplasmic components by a lysosomal membrane. This membrane has distinct functions, including the formation of a degradation-resistant barrier that protects the cellular structures against destruction, the generation of an acidic intralysosomal milieu, and the selective transport of degradation products to the cytoplasm. The biogenesis of new lysosomes during cell division and the turnover of lysosomal constituents require a continuous substitution with newly synthesized components. The targeting mechanisms and transport pathway of newly synthesized soluble hydrolases to lysosomes are better understood than those involved in targeting of lysosomal membrane proteins. Soluble hydrolases are equipped with mannose 6-phosphate (M6P) residues that are recognized by specific receptors mediating their transport from the Golgi to a prelysosomal compartment. The mechanisms of delivery of lysosomal membrane proteins are different and do not depend on M6P signals. Signal structures on the lysosomal proteins and their receptors as well as the identification of additional cytosolic and membrane-bound proteins involved in mediating the specificity of vesicular transport have been described in the last few years, as techniques for mutational cDNA analysis, in vitro assays, and analysis of immunocytochemical approaches have allowed the systematic molecular characterization of lysosomal targeting." @default.
